The 4* that first comes to mind is Commander Uhura. I think she's been available since the game began, but I've never seen her. Her stats are crap, but I'd still like to have her.
I actually keep a list of 4*s I don't have any copies of, and so here are the rest that I've never seen (including the voyage-exclusive Guinan and Worf). Also chasing stars on a number of others.

*I don't think this one is even in circulation yet via packs.
There is also Admiral Kirk, Delta Flyer Paris (which may no longer be an issue with next week's repeat event featuring him), Command Hologram (after getting him to 3/4 FE, he just vanished from drops), Mobile Doctor, Captain Scott, Crell Moset (I want to add more stars to mine), and Lt. Commander and Captain La Forges.
Oh, and Warship Chakotay. I have seen him not once!

Dr. Daystrom. Once had him show up in a behold, but chose other crew to get an extra star.
Guinan. Has not dropped in Gauntlet ever. Fortunately, Locutus did twice when he was there.
Disguised Kira. Same with Seven, never had a chance to get her. Wish I had for the upcoming event.
Wrathful Khan.
There are others, but those are my top five.
I'd also like more Data's to complete the collection, but not going to hold my breath.
I got four in the last month or so (two from voyages). Never saw him before then.
Defensive Phlox - after 7 opened packs he managed to avoid me pretty well.

Wrathful Khan. Dropped once, back in June or so. Haven't seen him since in hundreds of packs.
Crell Moset. Likewise. As useful as he is, I bought him a citation and will probably continue to do so. I've got him ff'd.
The Borg were the only characters I ever really wanted from this game, but I had the misfortune of starting to play right in the middle of the Borg mega event, so:
One and/or Borg Queen. Got both fe'd at 2/5. Would love to finish them someday. When the Rejoin the Collective pack came out, I was happy because it gave me a shot at these two plus Locutus... or so I thought. I must've bought fifteen of the stupid things and only got Drone Seven, Icheb, Drone Seven, Icheb, Drone Seven, Drone Seven, Drone Seven, Icheb... etc. None of the legendary Borg ever dropped for me. I strongly considered going FTP at that point.
Assimilated Janeway. Level 70 at 1/5.
Assimilated Torres eluded me for the longest time, but the other day I got her from the Zero Hour pack, then DYC'd her, then finished fusing her from more ZH packs -- and for once I only had to get maybe five packs total to get her ff'd. So I went from no Assimilated Torres to an immortal Assimilated Torres literally overnight.
Happy to see Delta Flyer Paris in next week's event. Got him ff'd at 2/5. I'm sure the other stars will elude me somehow, though.
Capt. Chaos I just got back into playing from a long hiatus and had Abraham Lincoln and Crell Moset drop in portals for me. I think I bought 5 portals and they were in there somewhere.
Lately, though, it's all been 3* rejects.
